Ο Schmidt (Jonah Hill) και ο Jenko (Channing Tatum) ήταν κάποτε συμμαθητές και καθόλου φίλοι. Ο πρώτος ήταν ο σπασίκλας και ο δεύτερος ο αθληταράς που τον κοπάναγε. Αρκετά χρόνια μετά, συμφιλιωμένοι, αποφοιτούν μαζί (και όπως-όπως) από τη σχολή της αστυνομίας και ανυπομονούν για να ριχτούν στην περιπέτεια της δουλειάς. Επειδή όμως δεν είναι και αυτό που λέμε "αστέρια", μετατίθενται άρον-άρον σε ένα παλιό, undercover πρόγραμμα της αστυνομίας. Πλέον, οι δύο συνάδελφοι θα δίνουν αναφορά στο τμήμα της 21 Jump Street και τον Capt. Dickson (Ice Cube), ενώ αποστολή τους θα είναι να γυρίσουν ξανά στα θρανία. Εκεί, θα πρέπει να παριστάνουν τα λυκειόπαιδα - κάτι που το φιζίκ τους το επιτρέπει, ενώ ως μυστικοί αστυνομικοί που είναι (;) καλούνται να εξαρθρώσουν ένα εξόχως επικίνδυνο και περίπλοκο κύκλωμα ναρκωτικών. Κι ενώ ο αυτοπεριορισμός δεν είναι το δυνατό τους σημείο, πρέπει εκτός από το έγκλημα να τα βάλουν και με ό,τι νόμιζαν πως είχαν αφήσει πίσω: την εφηβεία!
